ABSTRACT

Objective To explore the effects of nutritional intervention on the quality of life and nutritional status of patients with colorectal cancer treated with postoperative adjuvant chemotherapy. Methods Fifty-six patients with colorectal cancer subjected to postoperative chemotherapy were selected. They were divided into nutritional intervention group (n=20) and control group (n=36). Blood test indices, PG-SGA scores, and 36-Item Short Form Health Survey scores were compared 4 weeks after the surgery. Results No significant difference in PG-SGA score was found between the nutritional group and the control group at admission (t=-0.347, P=0.730), but statistically significant difference was detected after 4 weeks (t=-2.708, P=0.009). At 4 weeks after the surgery, the serum levels of prealbumin, albumin, and hemoglobin were not significantly different between the two groups (P > 0.05). Other indices of quality of life assessment (P < 0.05), except for the index of emotional function (P=0.083), were significantly different between the two groups, and the scores of the nutritional intervention group were higher than those of the control group. Conclusion Nutritional intervention can effectively improve the nutritional status and quality of life of patients with colorectal cancer treated with postoperative adjuvant chemotherapy.

ABSTRACT

ObjectiveTo predict the potential targets and mechanism of Jingfang mixture in the treatment of H1N1 influenza and provide references for clinical application of Jingfang mixture. MethodThe active components and targets of Jingfang mixture against H1N1 influenza were screened out by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),SwissTargetPrediction, and TargetNet. The targets of H1N1 influenza were obtained from GeneCards,Online Mendelian Inheritance in Man (OMIM), and DisGeNET and standardized by UniProt KB. The intersection targets were obtained by Venny 2.1.0. The "drug-component-target" network was constructed with Cytoscape 3.2.1 and analyzed for the topological attributes. The intersection targets were uploaded to STRING 11.5 to obtain the protein-protein interaction (PPI) network. Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis were carried out by Metascape. Finally,the top active components ranked by degree were docked to the core targets by Autodock vina and visually analyzed by PyMOL. Balb/c female rats were used for experimental verification. Hematoxylin-eosin(HE) staining was used to observe the pathological changes in lung tissues. Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of tumor necrosis factor-α(TNF-α),interleukin-10(IL-10), and interleukin-17(IL-17). Real-time fluorescence-based quantitative polymerase chain reaction(Real-time PCR) and Western blot were used to detect the mRNA and protein expression levels in lung tissues. ResultThere were 144 active components in Jingfang mixture. A total of 421 target genes of Jingfang mixture and 2 956 targets of H1N1 influenza were identified,including 199 common targets. Topological analysis showed that the core components of Jingfang mixture against H1N1 influenza included quercetin,luteolin, and kaempferol,and the core targets included prostaglandin-endoperoxide synthase 2(PTGS2),estrogen receptor alpha(ESR1),inducible nitric oxide synthase 2(iNOS2),peroxisome proliferator-activated receptorγ(PPARγ),and cyclooxygenase-1(PTGS1). GO enrichment yielded 697 items in biological process (BP) (P<0.01), 59 items in molecular function (MF)(P<0.01), and 21 items in cellular component (CC) (P<0.01). A total of 132 signaling pathways (P<0.01) were obtained by KEGG enrichment analysis, including phosphatidylinositol 3-kinases(PI3K)/protein kinase B(Akt) signaling pathway and mitogen-activated protein kinase(MAPK) signaling pathway,most of which were related to the regulation of immune inflammation. Molecular docking showed that the binding energy of the active components of Jingfang mixture to the core targets was less than -5.0 kcal·mol-1,indicating good binding activity. HE staining showed that the lung tissues were significantly improved after drug intervention,and Real-time PCR and Western blot showed that Jingfang mixture could reduce the mRNA and protein expression of PI3K and Akt in lung tissues. ConclusionJingfang mixture can play an anti-viral effect against the influenza A virus through multiple components,multiple targets, and multiple pathways. The active components quercetin,luteolin, and kaempferol may control the inflammation and regulate immunity on the PI3K/Akt,MAPK, and other signaling pathways by acting on targets such as PTGS2,ESR1,iNOS2,PPARγ, and PTGS1.

ABSTRACT

Borneol (Bo) and Arg-Gly-Asp (RGD) co-modified docetaxel (DTX) loaded MPEG-PLGA nanoparticles (DTX-Bo-RGD-NPs) were prepared to improve the therapeutic effect of DTX against glioma after intranasal administration. DTX-Bo-RGD-NPs were prepared by emulsification-solvent evaporation method, and their morphology, particle size, zeta potential, drug loading capacity (DLC), stability, and in vitro release properties were investigated. The fluorescence probe coumarin-6 loaded NPs were prepared for investigating the NPs' uptake property on C6 and 16HBE cell models to evaluate in vitro targeting ability. The DiR loaded NPs were prepared for observing the fluorescence intensity at the brain tumor site after intranasal administration through in vivo imaging system in a C6 rat orthotropic model, evaluating the targeting ability in vivo. The anti-tumor effects of DTX-Bo-RGD-NPs were also investigated in such C6 rat orthotropic model in vivo. Animal welfare and experimental procedures are in compliance with the regulations of the Animal Ethics Committee of Shanghai University of Traditional Chinese Medicine. The results showed that DTX-Bo-RGD-NPs were spherical and uniformly distributed, with a particle size of about 140 nm and a zeta potential of -20 to -30 mV. The drug delivery system showed good stability and sustained release property in vitro, and favorable brain tumor targeting effect in vitro and in vivo. Such novel drug delivery system significantly improved the accumulation of DTX-Bo-RGD-NPs in tumor sites and displayed a higher brain tumor targeting efficiency, providing promising therapeutics of DTX for the treatment of glioma after intranasal administration.

ABSTRACT

Objective:To investigate the correlation between endothelial dysfunction and cardiomyocyte apoptosis during myocardial ischemia-reperfusion.Methods:A total of 63 male rats were selected to establish the rat model of myocardial ischemia-reperfusion by the ligation of the left anterior descending(LAD)coronary artery to simulate myocardial ischemia.Rats were divided into the control group and group Ⅰb, group Ⅰa, group Ⅱb, group Ⅱa, group Ⅲb and group Ⅲa.Control rats were treated only with LAD threading without ligation.In observation group, at 30, 90, 120 min after LAD ligation(marked as Ⅰ, Ⅱ, Ⅲ group respectively), loosen the ligation to simulate ischemia-reperfusion.In the observation group, captopril sublingual injection of 0.25 mg/kg before ligation were marked as group b, and as group a with no captopril injection.The circulating endothelial cells(CEC), endothelin(ET), nitric oxide(NO)and the apoptosis rate of cardiomyocytes in each group were measured.Results:The CEC and ET levels showed a continuous upward trend, and a NO level showed a continuous downward trend from group Ⅰa to Ⅱa to Ⅲa as compared with the control group( P<0.05). After using preventive intervention of captopril, the CEC and ET levels were lower and NO levels were higher in group Ⅱb and Ⅲb than in group Ⅱa and Ⅲa, respectively( P<0.05). The apoptotic rate of cardiomyocytes was higher in group Ⅰa than in the control group, and the apoptotic rate from high to low were from group Ⅲa[(235.71±40.25)%]to group Ⅱa[(197.28±43.56)%]to group Ⅰa[(138.55±32.87)%]and to the control group[(5.81±2.02)%]( P<0.05). The apoptotic rate of cardiomyocytes was lower in group Ⅱb[(125.67±26.51)%]and Ⅲb[(124.91±33.28)%]than in group Ⅱa and Ⅲa, respectively( P<0.05). Conclusions:The ischemia-reperfusion can cause endothelial dysfunction and the apoptosis of cardiomyocytes, and there is a close relationship between the degree of this lesions and the duration of ischemia-reperfusion.While, the appropriate application of angiotensin converting enzyme inhibitor can inhibit the damage of cardiomyocytes to a some extent.

ABSTRACT

Objective To identify the risk factors of early post-surgical recurrence of hepatocellular carcinoma (HCC) within 2 years.Methods This retrospective study included 178 consecutive patients with HCC who underwent curative resection between January 2009 to December 2012 at Zhongshan Hospital,Fudan University.There were 151 males and 27 females,with a mean age of (58±11) years.The CT features including rim enhancement,satellite nodule,two-trait predictor of venous invasion (TTPVI),and nonsmooth tumor margins were reviewed.After hospital discharge,the patients were followed-up regularly for at least 2 years to detect tumor recurrence.The primary end point was recurrence of HCC.Results On univariate analyses AFP ≥ 200 μg/L,rim enhancement,TTPVI,non-smooth tumor margins and largest diameter >5 cm were correlated with early post-surgical recurrence of HCC.On multivariate analyses,AFP≥200 pg/L (HR=2.144,95%CI:1.350~ 3.406),rim enhancement (HR =2.196,95% CI:1.345 ~ 3.587),TTPVI (HR=1.735,95%CI:1.086~2.772),and non-smooth tumor margins (HR=2.065,95%CI:1.242~3.432) were independent risk factors of early post-surgical recurrence of HCC.Conclusion AFP≥200 μg/L,rim enhancement,TTPVI,and non-smooth tumor margins were independent risk factors of early post-surgical recurrence of HCC.

ABSTRACT

Natural antisense transcripts (NAT) and alternative polyadenylation (APA) of messenger RNA (mRNA) are important contributors of transcriptome complexity, each playing a critical role in multiple biological processes. However, whether they have crosstalk and function collaboratively is unclear. We discovered that APA enriched in human sense-antisense (S-AS) gene pairs, and finally focused on RNASEH2C-KAT5 S-AS pair for further study. In cis but not in trans over-expression of the antisense KAT5 gene promoted the usage of distal polyA (pA) site in sense gene RNASEH2C, which generated longer 3' untranslated region (3'UTR) and produced less protein, accompanying with slowed cell growth. Mechanistically, elevated Pol II occupancy coupled with SRSF3 could explain the higher usage of distal pA site. Finally, NAT-mediated downregulation of sense gene's protein level in RNASEH2C-KAT5 pair was specific for human rather than mouse, which lacks the distal pA site of RNASEH2C. We provided the first evidence to support that certain gene affected phenotype may not by the protein of its own, but by affecting the expression of its overlapped gene through APA, implying an unexpected view for understanding the link between genotype and phenotype.

ABSTRACT

Activating blood and dissolving stasis is one of the important principles of clinical anti-tumor by traditional Chinese medicine and the anti-tumor effect of the blood-activating and stasis-resolving drugs has been confirmed by a large number of studies. But in recent years, some experimental studies have found that some blood-activating and stasis-resolving drugs could promote tumor metastasis. And then the clinical application of this kind of drugs has suffered from many controversial problems. Chinese materia medica (CMM) with breaking blood stasis and resolving mass (Poxue herbs) is a kind of blood-activating and stasis-resolving drugs which display the severe effect. Nowadays some researchers confirmed its obvious antineoplastic effect and its prominent anti-tumor metastasis effect, which indicated in recent studies. In this article, we summarized the anti-tumor metastasis effect and mechanism of the Poxue herbs like Curcumae Rhizoma, Sparganii Rhizoma, large blister Beetle, leech, and its effective components in detail. And we also put forward that Poxue herbs are the ones which have the anti-tumor metastasis effect among blood-activating and stasis-resolving drugs, and it valued with significant clinical application and development. It is also believed that the Poxue herbs could inhibit the tumor metastasis which is related with aerobic glycolysis and can provide the theoretical direction to guide clinical application.

ABSTRACT

Circular RNAs (circRNAs), a kind of covalently closed RNA molecule, were used to be considered a type of by-products of mis-splicing events and were discovered sporadically due to the technological limits in the early years. With the great technological progress such as high-throughput next-generation sequencing, numerous circRNAs have recently been detected in many species. CircRNAs were expressed in a spatio-temporally specific manner, suggesting their regulatory functional potentials were overlooked previously. Intriguingly, some circRNAs were indeed found with critical physiological functions in certain circumstances. CircRNAs have a more stable molecular structure that can resist to exoribonuclease comparing to those linear ones, and their molecular functions include microRNA sponge, regulatory roles in transcription, mRNA traps that compete with linear splicing, templates for translation and possibly other presently unknown roles. Here, we review the discovery and characterization of circRNAs, the origination and formation mechanism, the physiological functions and the molecular roles, along with the methods for detection of circRNAs. We further look into the future and propose key questions to be answered for these magical RNA molecules.

ABSTRACT

The breeding of domestic rabbits (Oryctolagus cuniculus) for human consumption has a long tradition in China. Infections that can affect the production of meat or even be transmitted from animals to humans are important to monitor, especially for public health reasons as well as for their impact on animal health. Thus, a total of 1,132 domestic rabbit sera from 4 regions in China were collected for serological screening for Encephalitozoon cuniculi and for Toxoplasma gondii by ELISA and modified agglutination test (MAT), respectively. Antibodies to E. cuniculi were detected in 248/1,132 (21.9%) sera tested while antibodies against T. gondii revealed a seroprevalence of 51/1,132 (4.5%). We believe that the present results are of epidemiological implications and public health importance due to the acknowledged susceptibility of humans to E. cuniculi and T. gondii infections. Therefore, routine screening tests of domestic rabbits are proposed considering the zoonotic potential of these parasites.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of hesperidin on behavior and hypothalamic-pituitary-adrenal (HPA) axis of ratmodel of chronic stress-induced depression.</p><p><b>METHOD</b>Chronic unpredictable mild stress (CUMS) was used to establish the rat depression model. Sixty male SD rats were divided randomly into six groups: the normal group, the model group, the hesperidin (40, 80, 160 mg x kg(-1)) group and the positive fluoxetine (10 mg x kg(-1)) group. They were orally administered with drugs for three weeks. The sucrose preference test and the forced swimming test (FST) were assayed to detect animal behavior. The levels of corticosterone (CORT) in serum, mRNA of corticotropin release factor (CRF) in hypothalamus as well as protein expression of glucocorticoid receptor (GR) in paraventricular nucleus (PVN) were determined to clarify the anti-depression effect and mechanism of hesperidin.</p><p><b>RESULT</b>Compared with the model group, rats in the hesperidin (40, 80, 160 mg x kg(-1)) treatment group showed significant increase in the sucrose consumption and decrease in the immobility time in FST to varying degrees. Meanwhile, the excessively high serum CORT and adrenal index of CUMS rats were reversed by treatment with hesperidin. In addition, hesperidin inhibited CRF mRNA expression in hypothalamus and up-regulated GR protein expression in PVN among CUMS rats.</p><p><b>CONCLUSION</b>Hesperidin could effectively improve the behavior of CUMS rats and show the anti-depression effect. Its mechanisms may be related to the function of regulating HPA axis.</p>

ABSTRACT

Objective: To develop a novel method using high-performance capillary electrophoresis coupled with DAD (HPCE-DAD) for the simultaneous determination of 10 phenolic compounds (luteolin-7-O-glucoside, rutin, quercetin-3-glucoside, quercetin-3- rhamnoside, salicylic acid, luteolin, kaemferol, quercetin, gallic acid, and protocatechuic acid) in Bidens bipinnata. The extraction efficiency of above mentioned compounds was compared using six different ethanol/water solutions. Methods: The effects of pH value and concentration of buffer, applied voltage, and temperature on the separation were researched. The 10 compounds were baseline separated within 16 min in a 60 cm length capillary at a separation voltage of 25 kV with a running buffer consisting of 25 mmol/L borate (pH 9.5) at wavelength of 214 nm. Results: Excellent linearities of luteolin-7-O-glucoside, rutin, quercetin-3-glucoside, quercetin-3-rhamnoside, salicylic acid, luteolin, kaemferol, quercetin, gallic acid, and protocatechuic acid were observed at 5.0-120, 5.0-120, 5.0-120, 5.0-120, 1.0-120, 2.5-120, 5.0-120, 2.5-120, 2.5-120, and 2.5-120 mg/L, respectively. The detection limits were at 2.5, 2.5, 2.5, 2.5, 0.5, 1.0, 2.5, 1.0, 1.0, and 1.0 mg/L, respectively. The relative standard deviations (RSD) of precision were below 5.17% (n = 6). The mean recoveries for 10 phenolic compounds in B. bipinnata ranged from 94.4% to 105.8%, with RSD values of 0.32%-4.33%. Conclusion: The contents of luteolin-7-O-glucoside, rutin, quercetin-3- glucoside, quercetin-3-rhamnoside, salicylic acid, luteolin, kaemferol, quercetin, gallic acid, and protocatechuic acid in B. bipinnata are 95.04-105.84, 78.03-110.45, 96.45-177.96, 178.78-300.00, 62.06-66.54, 71.08-72.85, 77.39-95.30, 68.27-77.43, 68.47- 88.47, and 107.24-142.43 μg/g, respectively.

ABSTRACT

Objective To observe the influence of intensive control of blood glucose, blood pressure and blood lipid on fasting plasma obestatin level in type 2 diabetes mellitus (T2DM) patients and its correlation with lipid metabolism. Methods Seventy-two T2DM patients were assigned to four groups according to different treatments. Group A(n=25) received blood glucose (BG) and blood pressure(BP)controlling; group B(n=12) received BG, BP and blood lipid (BL) controlling; group C(n=20) received BG,BP, and BL controlling plus vitamin E administration; and group D(n=15) received BG,BP,BL controlling and compound Danshen dripping pill administration. Results Fasting obestatin level of group A was significantly lower than those in the other three groups (P<0.01). Fasting obestatin level was negatively correlated with body mass index (BMI), HbAlc, waist to hip ratio (WHR),total cholesterol(TC), low-density lipoprotein cholesterol (LDL-C), fasting insulin (FIN) and hemeostasis model assessment of insulin resistance (HOMA-IR) (P<0.05), and was positively correlated with high-density lipoprotein cholesterol (HDL-C) (P<0.05). After adjusting age, sex, BMI, WHR, blood glucose, and blood pressure, high level fasting obestatin was correlated with intensified control of blood lipid (P<0.01). Conclusion The fasting obestatin level is increased in T2DM patients receiving BL control compared with those do not receive, and obestatin level might be associated with lipid metabolism.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of adipose stromal vascular fraction cells (SVFs) with VEGF on the neovascularization of free fat transplantation.</p><p><b>METHODS</b>SVFs were obtained from subcutaneous fat and labelled with DiI. 0.3 ml autologous fat tissue was mixed with 0.2 ml cells: 1) autologous SVFs with VEGF (Group A); 2) autologous SVFs (Group B); 3) complete DMEM (Group C) And then the mixture was injected randomly under the back skin of 12 nude mice. The transplanted fat tissue in three groups was harvested at 2 months after implantation. Wet weight and diameter of fat grafts was measured. After HE and CD31 staining,blood vessel density, viable adipocytes and fibrous proliferation were observed.</p><p><b>RESULTS</b>Trace of SVFs labeled by DiI in vivo could be detected by fluorescent microscope. The wet weight of fat grafts was (191.90 +/- 9.81) mg in group A, (177.01 +/- 10.50) mg in group B, and (92.05 +/- 8.30) mg in group C (P<0.01). The diameter of fat grafts was (0.49 +/- 0.24) cm in group A, (0.40 +/- 0.26) cm in group B, and (0.32 +/- 0.28) cm in group C (P<0.01). Histological analysis showed the blood vessel density was (14.58 +/- 2.06)/HPL in group A, (11.55 +/- 2.18)/HPL in group B, (7.87 +/- 1.55)/HPL in group C. Compared with group B and group C, group A had more adipose tissue with less fat necrosis and fibrosis and had significantly higher capillary density.</p><p><b>CONCLUSIONS</b>The autologous adipose stromal vascular fraction cells with VEGF could improve the neovascularization of free fat significantly. It indicates a wide clinical application in the future.</p>
